Proposed model for Shh-GLI signaling-mediated proliferation of mantle cell lymphoma (MCL): targeting GLI in hedgehog pathway leads to improved chemosensitivity: activation of hedgehog signaling (Shh-GLI signaling) in MCL cells leads to over-expression of GLI transcription factors. These over-expressed molecules in turn induce the expression of cyclin D1 and BCL2, which thereby increase proliferation/survival and resistance to therapy in MCL. When overexpressed GLI is downregulated by GLI-specific antisense oligonucleotides, GLI-mediated cyclin D1 and BCL2 expression is decreased, which leads to decreased proliferation and increased susceptibility of MCL to therapy. “?” symbol indicates that the constitutive activation of the signaling molecules may be due to mutations in the molecules.
