Tumor-bearing animals received the pH-sensitive dye acridine orange via their tail vein, their lungs were excised 2 hours later and examined for acidity in the metastases by fluorescence stereoscopic microscopy. B16-F10 melanoma cells in lung metastases showed extensive accumulation of acridine orange (green), suggesting that the cells created an acidic microenvironment.
