Long noncoding RNAs (lncRNAs) exhibit a myriad oncogenic functions. In their study on page 746, Shao and colleagues performed transcriptomic analysis using The Cancer Genome Atlas to search for dysregulated lncRNAs in lung adenocarcinoma (LUAD). The authors discovered increased expression of lncRNA MSL3P1 in LUAD, and found MSL3P1 negatively correlated with overall survival in LUAD patients. Epigenomic analysis using public data revealed enhanced MSL3P1 expression in LUAD may be mediated by an interaction between an aberrantly-activated enhancer and the MSL3P1 promoter. The cover features a Hi-C matrix visualizing genomic MSL3P1 promoter interactions, where the top matrix depicts interactions in a LUAD cell line and the bottom matrix presents interactions in normal lung tissue. Subsequent experiments demonstrated that MSL3P1 enhanced expression of E3 ubiquitin ligase cullin 3 by disrupting degradation of cullin 3-encoding CUL3 mRNA by ZHC3H1. The MSL3P1-CUL3 axis promoted LUAD cell invasion and migration in vitro and metastasis in a mouse model, identifying a novel mechanism by which lncRNA facilitates metastasis in LUAD. - PDF Icon PDF LinkTable of Contents
