AKT phosphorylates FOXO, which causes shuttling of FOXO1 from nucleus to cytoplasm. Cytoplasmic localization of FOXO1 protects the cells from FOXO/Bim-induced apoptosis. Treatment of BxPC-3 cells with benzyl isothiocyanate (BITC) induces nuclear retention of FOXO1 resulting in the transcriptional upregulation of Bim, leading to apoptosis in pancreatic cancer cells. BxPC-3 cells were treated with 10mmol/L BITC for 24 hours and immunostained with FOXO1 (red) and actin (green) antibodies overnight, followed by a 2-hour incubation with Alexa fluor 594 and Alexa fluor 488 antibodies, respectively. Cells were photographed under microscope (60x) after staining the nucleus with DAPI (blue).
