Introduction: Daucus carota, Apiaceae, commonly used in Lebanese folklore medicine for the treatment of leukemia. The anti-cancer aqueous extract of DC was examined by evaluating its effect on the induction of differentiation and inhibition of proliferation of the human promyelocytic leukemia HL-60 cells.Materials and Methods: The effects of non-cytotoxic concentrations of DC were evaluated against HL-60 cells. The effect of DC on the proliferation was evaluated by MTT assay and TGF mRNA protein expression using RT-PCR. The anti-proliferative effects were tested by determining the changes in the expression of transforming growth factors (TGF-alpha, TGF-beta1 and TGF-beta2). In parallel, the differentiation effect was measured by NBT reduction assay. The degree of differentiation was assayed by the ability of Results: DC was found to reduce cell proliferation and induced cell differentiation. Conclusion: The results of this study show DC extract possesses anti-proliferative effects and stimulates of differentiation of HL-60 cells.
