Major Finding: BRCA2 protects RAD51 stability at DNA repair sites by hindering the retention of PARP1 on DNA.
Concept: RAD51-destabilizing PARP1:DNA binding is enhanced in BRCA2-deficient cells following PARP inhibition.
Impact: This study provides a mechanism for heightened response to PARP inhibition in BRCA2-mutant cancers.
