Ligand Engagement Induces LAG3 Checkpoint Activity through Ubiquitination

Major Finding: Ligand engagement–induced polyubiquitination activates LAG3 by blocking LAG3 membrane binding.
Concept: LAG3 ubiquitination is mediated by Cbl family ligases and is required for LAG3 checkpoint function.
Impact: Coexpression of LAG3 and CBL is a potential biomarker for response to LAG3-targeted immunotherapy.
