Major Finding: AARS1 mediates protein lactylation that couples cell metabolism and p53 tumor suppressor function.

Concept: AARS1 lactylates p53 at lysines 120 and 139 to block DNA binding and transcriptional activation.

Impact: Inhibiting p53 lactylation via β-alanine treatment may benefit patients with p53 wild-type cancers.
