PGE2 Disrupts IL2 Signaling to Limit Tumor-Infiltrating Lymphocyte Expansion

Major Finding: PGE2 inhibits stem-like CD8+ tumor-infiltrating lymphocyte (TIL) expansion and differentiation by disrupting IL2 signaling and induces TIL death via oxidative cell stress and ferroptosis.

Concept: IL2 sensing is limited by signaling through the PGE2-EP2/EP4 axis, and inhibition of this signaling in TILs restores tumor control.

Impact: These results suggest that targeting the PGE2-EP2/EP4 axis in TIL could enhance cancer immunotherapy efficacy.
