Purpose:We studied the effect of feiyanning decoction on NF-kappaB activated by TNF-\#945; in lung adenocarcinoma cells (A 549). Methods:A549 cells were incubated with rat serum containing feiyanning decoction at different concentrations for 24, 48 h, respectively. Morphology of cells was carried out after treatment with reagents. The cell proliferation was examined by the WST-8 assay. The NF-kappaB and I\#954;B\#945; expressions were studied by western blotting. The transcriptional activity of NF-kappaB promoter was detected by Luciferase assay. Results: TNF-\#945; (1 ng/ml) strongly induced the expression of NF-kappaB by approximately 1.76-fold compared with the control in the nuclei of A 549 cells, the induced NF-kappaB expression was significantly suppressed by the addition of feiyanning (p < 0.01). In addition, Feiyanninginhibitedthetranscriptional activity of theNF-kappaB promoter. However, we observed no significant changes in I\#954;B\#945; expression (p > 0.05). Conclusion: These results suggest that feiyanning decoction inhibits NF-kappaB activation induced by TNF-\#945;. It is therefore expected to play a new strategy to treat lung cancer.
